Isolation of high-quality RNA from the tissues of perennial woody plants, including woody grape vines, is very difficult due to the high content of phenolic compounds, secondary metabolites and polysaccharides and the ribonuclease activity of destroyed tissues. Most of the existing methods require either large time or financial costs, or do not give reproducible results in the case of RNA extraction from mature grape tissues. The modified isolation protocol is based on a combination and modification of the known RNA extraction methods, taking into account the characteristics of mature grape tissues. Existing commercial kits for the isolation of RNA from plant tissues showed a low efficiency of RNA extraction from mature grape tissues, primarily associated with "varietal specificity". Reproducible results in the extraction of RNA showed CTAB-method, however, it has several significant drawbacks associated with the duration of the extraction and the complexity of the processing of an RNA preparation with a DNAase. The developed method is based on increasing the concentration of mercaptoethanol and polyvinylpyrrolidone in the extraction buffer, eliminating the stage of RNA selective precipitation via LiCl, and replacing it with deposition on a silica-based membrane (SiO2) followed by processing with DNA-ase. and increase the purity of the preparation of RNA from genomic DNA in comparison with the original method. A modified isolation protocol was developed based on a combination and modification of known RNA extraction methods, taking into account the characteristics of mature grape tissues. This solution allows to obtain reproducible quantity and quality of RNA for the subsequent synthesis of cDNA and RT-PCR
The article is devoted to process of the state
development and emergence of new forms of the
government in the region of the North Caucasus during
the Civil War. The author tells how forms of a political
system and the government in the region after October
revolution of 1917 have changed. He analyzes the
process of emergence of the new military-political
modes and character of a state system in its various
areas. In the article, the political background of
appearance of new forms of statehood and power
organization is also considered. The author equally
investigates the forms of the state life created by
Bolsheviks and their supporters, and the forms of their
political opponents – representatives of the white, antiBolshevist
movement. The author emphasizes that it was
the period of disintegration of traditional Russian
statehood and traditional forms of the organization of the
political power in the region. New options of the state
system which are directly connected with the crisis of
the Russian society in 1917 y. came in the region to
replace the old ones. In some cases, they represented
rather non-standard forms of the organization of the
government, including formation of the Soviet republics
in the territory of the region, appearance of the ataman
power and military dictatorships. On the one hand, they
were based on ethnocultural specifics of regional life,
and on the other hand they were defined by the crisis of
the period of the Civil War
Under contemporary conditions in Russia, patriotism
took again its rightful place in the treasury of the
education of the younger generation. Having gone
through many years of testing, attempts to oblivion,
libel and slander, patriotism has been returned to
pedagogical arsenal by efforts of the best Russian
people. However, the atmosphere of decelerations and
partization, nationalist sentiment increasing, immoral
behavior of young people are persisting. Percentage of
maritime university’s cadets has a declining interest in
the patriotism subject. Thus, 78% of cadets reported
that they did not read fiction on patriotic themes; 53%
do not watch military-patriotic films; 29% are not
interested in the history of his native city. The data
indicates a lack of using educational potential and the
special flavor of the hero cities, where they are located,
their rich and glorious history, and established for ages
tradition of serving the motherland. Now for this
reason, the administration, the teaching staff of
maritime universities have not only an intention of
qualitive patriotic education, but also to search for new
forms and methods, to use a regional and municipal
component, to develop cadet’s attention, interest and
desire to participate in patriotic events and patriotic
activities. The working on cadet’s patriotic education of
maritime university can become one of the most
modern and innovative directions. This work shall
consider conditions, historical and educational potential
of the city, where an educational institution is located,
studying of its glorious history, combat and labor
traditions, as the cognitive basis for the patriotism
formation of the highest school’s students

The created variety is a complex inter-species Euro-American-Amur hybrid. The duration of the production period is 146-155 days. The growth of shoots is strong. The degree of grapevine maturing high. It is stainable for high winter hardiness, drought resistance and increased resistance to fungal diseases, tolerant to phylloxera. The top of the young shoot is green without pubescence. Long-term storage of fruits can be achieved by means
of decreasing microbial contamination of their surface
as a result of fruits pre-treatment by biopreparations
before dispatching for storage. In the current study
patterns of influence of fruits pre-treatment with the
“Ekstrasol” biopreparation in a number of
concentrations of bioagent on the microbial
contamination of their surface during storage are
revealed. The “Ekstrasol” preparation is a biofungicide,
containing the Ch-13 strain of Bacillus subtilis bacteria
and their metabolites. As research objects there were
chosen apples of a zoned in the Krasnodar region
variety called Idared and pears of a zoned in the
Krasnodar region variety called Conference, harvested
in 2016. The apples and the pears were stored over a
period of 6 months in the low temperature conditions.
Samples were withdrawn every month over the period
of storing. The species composition and quantity of the
most prevalent microorganisms, potentially causing the
spoilage of apples and pears during storing, were
identified. The optimal concentration of the bioagent,
which provides the maximal decreasing of microbial
contamination of fruits’ (apples and pears) surface, is
established – 106
CFU/g, dosage of solution – 1 ml per 100 g of fruit

This article analyzes the representations of modern
youth of heroism as the basis of patriotic and civic
education. Special attention is paid to consideration of
the transformation of the concept of "heroism", reduce
the social significance of the feat in the consciousness
of modern Russian society. Value of creating an image
of the hero as a way of formation a sense of patriotism
and learning patterns of social activity necessary for
the creation of a civil society is analyzed. On the basis
of sociological research among students of higher
education institutions of Krasnodar, the understanding
of the essence of "heroism" and attitude to the heroic
deeds is revealed. The connection of this social
phenomenon with a set of personal lines and moral
qualities of the person is established. The most
favorable for commission of feats spheres and the
personalities applying for a role of heroes for today's
younger generation are defined. The authors draw a
conclusion about the existence of student’s interest in
the phenomenon of heroism and the passive need for
the formation of a distinct image of the hero on a basis,
both the celebrated historic figures, and
contemporaries. In addition, discrepancy in a number
of answers of students is noted, indicating the absence
of a modern young people a clear idea of heroism and
heroic acts, their importance in personal development.

productivity in the agrarian sector of Russia and the

trends and factors affecting the growth of this indicator
were revealed. One the main reasons for low

achieved by improving the employment and
organizational-technological innovation. We have
comprehensively illuminated the problem of labor
motivation. Based on our own researches and data
from agrarian scientists the ways of its strengthening
were traced. Also, there was highlighted highly
motivated work in the family farms, which is not just a
form of entrepreneurial employment based on private
ownership of the means of production and the use of
familial form of labor, but also the lifestyle of the rural
man. We must not forget that the main motivation to
work lies not only in its pay, but also in the education
of people and traditions. It highlights the role of social
support of rural youth and the need for radical
improvement of social infrastructure. For the selfassertion
of the individual, which is especially
important for young men, the person should see a
sense of ownership, initiative, independence,
participation in decision-making and the distribution of
income. These qualities are fully manifested in the
